Terrestrial planarians (Platyhelminthes, Tricladida, Terricola) from the Iberian peninsula: first records of the family Rhynchodemidae, with the description of a new Microplana species Naturalis

The first records of terrestrial planarians belonging to the family Rhynchodemidae are reported for the Iberian Peninsula. A new endemic species from the Spanish Pyrenees, Microplana nana sp. nov., is described. The characteristic features of this species are: i) small size (8-10 mm) in adult individuals, ii) very long conical penis papilla and iii) absence of seminal vesicle, bursa copulatrix, genito-intestinal duct, and well-developed penial bulb. The solitary egg parasitoids (Hymenoptera: Scelionidae) of Pentatomidae species complex are potential candidates for soybean (Glycine max L.) integrated pest management programs in Brazil. The correct identification of species and strains of these parasitoids is necessary to implement the use of biological control. The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters. The program RAPDDIP applied to S. pallens revealed nucleotide diversity of 2.3 and 2.2% for the populations from Rio Grande do Norte (Northeast Brazil) and Federal District (Central Brazil), respectively. Random amplified polymorphisms between two South American subspecies of rattlesnakes (Crotalus durissus collilineatus and Crotalus durissus terrificus) BABT

The present work was made to determine the suitability of RAPD analysis for the identification of specimens of two subspecies of South American rattlesnakes (Crotalus durissus terrificus and Crotalus durissus collilineatus). The 11 arbitrary primer sequence tested amplified a total of 161 bands, of which 31% were polymorphics when compared with Crotalus specimens. The combining results from different primers allowed the identification of all the specimens under analysis. Several characteristic bands of each subspecies were identified. Dendrogram, based on RAPD markers, show three groups that corresponded to the subspecies of Crotalus, and to Bothrops jararaca specimen.

The soybean (Glycine max (L.) Merrill) frogeye leaf spot is caused by the fungus Cercospora sojina Hara and is a widespread disease in Brazil and other countries, causing severe losses in grain yield and also affecting seed quality. The availability of DNA markers linked to genes for resistance to this disease would accelerate breeding programs, particularly when other traits are also being evaluated. Data from five protein-coding loci related to dairy production were used to study the genetic diversity and population structure of Argentine and Bolivian Creole cattle breeds. Microsatellite markers or SSR (Simple Sequence Repeats) have proved to be an excellent tool for cultivar identification, pedigree analysis and the evaluation of genetic distance among organisms. Soybean cultivars have been characterized mainly by morphological and biochemical traits. However, these traits have not been sufficient to characterize the large number of cultivars eligible to receive protection under the Brazilian Cultivar Protection Act. Bemisia tabaci (Genn.) was considered a secondary pest in Brazil until 1990, despite being an efficient geminivirus vector in beans and soybean. In 1991, a new biotype, known as B. tabaci B biotype (=B. argentifolii) was detected attacking weed plants and causing phytotoxic problems in Cucurbitaceae. Nowadays, B. tabaci is considered one of the most damaging whitefly pests in agricultural systems worldwide that transmits more than 60 different plant viruses. Amplified fragment length polymorphism (AFLP) is a fast and reliable tool to generate a large number of DNA markers. In two unrelated F2 populations of carrot (Daucus carota L.), Brasilia x HCM and B493 x QAL (wild carrot), it was hypothesized that DNA 1) digested with the same restriction endonuclease enzymes and amplified with the same primer combination and 2) sharing the same position in polyacrylamide gels should be conserved sequences. The direct amplification of length polymorphism technique (DALP) has been used to distinguish species-specific banding patterns in two marine gastropod oyster drills Ocenebra erinacea (Linnaeus, 1758) and Ocinebrellus inornatus (Recluz, 1851). Ocenebra erinacea is the European oyster drill, common along all European coasts. Ocinebrellus inornatus, the Japanese oyster drill, was recorded in oyster growing areas of the Marennes-Oleron Bay (SW France) for the first time in 1995. Sequence-Characterized Amplified Region (SCAR) appears as a useful technique for genetic purity testing and variety discrimination, applicable to species in which some other techniques have failed. In particular, this technique is very attractive with species in which RAPD results were not consistent. The RAPD polymorphic bands were cloned, sequenced and from the sequence information, primers pairs for normal PCR were developed. Since the probability of obtaining successful SCAR primers from RAPD polymorphic bands was about 50%, a larger number of RAPD polymorphic bands are needed to develop sufficient SCAR primers for varietal discrimination in vinca. Although the first inhabitants of western Amazonia domesticated pejibaye (Bactris gasipaes Kunth, Palmae) or peach palm for its fruits, today it is widely planted for its heart-of-palm. Like other domesticates, pejibaye presents a complex hierarchy of landraces developed before the conquest of the Americas. The existence of three landraces (Pará, Solimões, Putumayo) was proposed along the Amazonas and Solimões Rivers, Brazil, based on morphological characteristics. There are some questions remaining about the intermediate landrace being an artifact of the morphometric analysis. The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis. Initially, the two parental DNAs from the resistant donor SC09 and 'Araguaia' were analyzed using random primers. Of the 240 primers tested, 203 produced amplification products. An epidemic of rice (Oryza sativa) blast occurred on cultivars Epagri 108 and 109 in the municipalities of Lagoa da Confusão and Duerê in the State of Tocantins, during the rice-growing season 1998-99. DNA fingerprinting and virulence phenotype analysis were utilized to determine the diversity of Pyricularia grisea isolates collected from these cultivars in one epidemic year. Total protein electrophoresis and RAPD fingerprinting analysis for the identification of Aeromonas at the species level BJM

Fifteen well-defined strains of Aeromonas of thirteen species were analyzed by SDS protein electrophoretic analysis (SDS-PAGE) and random amplified polymorphic DNA analysis (RAPD). The comparison between the patterns obtained by both methods allowed differentiating all the strains. Clusters formed by the unweighted pair group method with arithmetic averages applied to protein data correlates with the genetic and biochemical information about the species. The results show that protein fingerprinting has the potential to differentiate Aeromonas species, but the low qualitative variation indicates that this technique is not efficient for the characterization of strains within a species. Reciprocal recurrent selection effects on the genetic structure of tropical maize populations assessed at microsatellite loci Genet. Mol. Biol.

A modified reciprocal recurrent selection (RRS) method, which employed one cycle of high-intensity selection, was applied to two tropical maize (Zea mays L.) populations, BR-105 and BR-106, originating the improved synthetics IG-3 and IG-4, respectively. In the present study the effects of this kind of selection on the genetic structure of these populations and their synthetics were investigated at 30 microsatellite (SSR) loci. A total of 125 alleles were revealed. A reduction in the number of alleles was observed after selection, as well as changes in allele frequencies.
